The suburb derives its name from William Redfern, a surgeon granted 100 acres (0.40 km²) of land in 1817 by Governor Lachlan Macquarie. Redfern built a country house surrounded by vibrant flower and kitchen gardens. Prominent neighbors included Captain Cleveland, who constructed Cleveland House, and John Baptist, renowned for his nursery and seed business.
The area's development gained momentum with the establishment of Sydney’s first railway terminus in Cleveland Paddocks, extending from Cleveland Street to Devonshire Street. This terminus honored William Redfern, although the current Redfern railway station was initially named Eveleigh. When Central Station was later constructed, the original terminus site became Prince Alfred Park.

Redfern was incorporated as a borough in August 1859 and later merged with the City of Sydney in 1949.
